Will this work for a 4th gen 4runner w/ 3rd row option?

The floor is is not compatible with 3rd row equipped 4Runners.

Will this floor system work with other sliders and drawers? I’m looking at getting the another Slide and fridge. Was wanting to make sure this is compatible with it?

You may run into issues with other brands bolt patterns being different, but you can physically mount just about anything to the floor since its just a flat surface. Other brands products however, may require some modifications.
